Phys­ioS­tim: car­diac safety pharmacology

Since 2000, Phys­ioS­tim devel­oped elec­tro­phys­i­o­log­i­cal mod­els to inves­ti­gate the poten­tial of com­pounds to induce car­diac adverse effects.

Our mod­els are based on three techniques:

  • Intra­cel­lu­lar micro­elec­trode assays: action poten­tial recordings
  • Patch clamp in whole cell or per­fo­rated patch con­fig­u­ra­tion: human ion chan­nel assays
  • Iso­lated Lan­gen­dorff per­fused heart: ECG and haemo­dy­namic parameters
  • Iso­lated organ bath: contractility

The design per­formed for a non GLP study (=“GLP spirit”) is basi­cally the same except for the num­ber of car­ried exper­i­ments, the con­cen­tra­tions tested (accord­ing to the Spon­sor), whether a vehi­cle group is used or not and whether for­mu­la­tion of test arti­cle solu­tions per­formed or not.
